Product Specific Information

The monoclonal antibody IM348 recognizes mouse CD6, a transmembrane glycoprotein of the scavenger receptor family. The extracellular domain contains three scavenger receptor cysteine-rich (SRCR) domains, a transmembrane region, and a cytoplasmic domain of variable length depending on the isoform. CD6 is expressed on most T lymphocytes. Expression differs slightly between human and mouse; in mice, splenic B cells lack CD6, while in humans, a subset of B lymphocytes express it.

The CD6 ligand is CD166 (Activated Leukocyte Cell Adhesion Molecule, ALCAM). Through interactions with CD166 or homotypic CD6-CD6 binding, CD6 plays a role in adhesion and migration during organogenesis. The intracellular domain of CD6 is constitutively phosphorylated, suggesting involvement in T cell activation and signaling. In humans, depletion of CD6-expressing cells increases graft-versus-host tolerance in transplants.

Applications Reported:
This IM348 antibody has been reported for use in flow cytometric analysis, immunoblotting (WB), and immunohistology staining of frozen tissue sections.

Applications Tested:
This antibody has been tested by flow cytometric analysis of mouse splenocytes and can be used at ≤0.25 µg per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be empirically determined (typically 10⁵–10⁸ cells/test). Antibody titration is recommended for optimal assay performance.

The IM348 antibody can also be used at <5 µg/mL in immunoblotting of non-reduced lysates.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

CD6, also known as T12, is a member of the scavenger receptor superfamily found on subsets of T and B cells, thymocytes, and acute lymphocytic leukemia (ALL) cells. CD6 interacts with its ligand CD166/ALCAM and serves as a coreceptor for T cell activation and stabilizer of the immunological synapse. CD6–ALCAM-mediated adhesion contributes to T cell proliferation. CD6 may also interact with CD5, modulating CD5 signaling. Ligation of CD6 has an anti-apoptotic role in chronic lymphocytic leukemia B cells.
