Target Information
The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
This tag is commonly expressed at either the N- or C-terminal regions of recombinant proteins to enable isolation or purification via immobilized metal affinity chromatography (IMAC).
Epitope tagging allows detection of recombinant proteins by fusing a known epitope to them.
His-tagged proteins can be purified using metal chelate chromatography due to the strong and selective binding affinity between histidine clusters and metal ions such as Ni²⁺.
His tag-specific antibodies are widely used for detection or co-immunoprecipitation of tagged proteins.
