Target Information
Complex V, also called F1F0 ATPase or ATP synthase, is highly conserved across species and found in plants, fungi, bacteria, and animals. It catalyzes the reversible reaction of ADP + phosphate to ATP during oxidative phosphorylation and functions as a proton-pumping ATPase.
Although primarily localized to mitochondria, ATP synthase has also been identified on plasma membranes of various cell types, such as hepatocytes (HDL receptor), endothelial cells (angiostatin receptor), and cancer cells.
The enzyme comprises two main sections: the soluble F1 ATPase (α, β, γ, δ, ε subunits) and the membrane-embedded F0 ATPase (a, b, c subunits). In mammals, ATP synthase consists of 17 subunits, five forming the F1 portion and the rest forming stalk domains and the proton-pumping F0 machinery.
Two F0 subunits are encoded by mitochondrial DNA, while others are nuclear-encoded. Mutations in mitochondrial-encoded subunits of ATP synthase (Complex V) are associated with OXPHOS disease.
