Product Specific Information

The 104 monoclonal antibody reacts with the mouse CD45 molecule, the leukocyte common antigen (LCA) in CD45.2-expressing mouse strains. These include C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR strains. CD45.2 is expressed by all leukocytes in these strains.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1 µg/test (in 100 µL final volume). Cell number can range from 10⁵ to 10⁸ cells/test. Optimal titration recommended.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the 405 nm violet laser and emits at 600 nm. Recommended detection filter: 610/20 bandpass. Ensure the instrument supports this fluorochrome.

Staining Buffer Recommendation:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored up to 3 days at 2–8°C in the dark using:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of surface proteins in lymphocytes. It regulates Src-family kinases, plays roles in lymphocyte development, antigen signaling, and integrin-mediated signal transduction. CD45.2 exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical cytoplasmic regions. These isoforms vary by cell type and physiological state. CD45RA is a related isoform with restricted expression among lymphoid subtypes.
