Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except mature erythrocytes and platelets. The cytoplasmic domain has tyrosine phosphatase activity essential for lymphocyte activation.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Mouse bone marrow cells, ≤0.25 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
• Fluorochrome Information: Super Bright 600 is excited at 405 nm and emits at 600 nm. Recommended filter: 610/20 bandpass.
• Staining Buffer: Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to reduce non-specific interactions.
• Light Sensitivity: Protect from light to prevent photo-induced oxidation.
• Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal impact on brightness.
• Laser Compatibility: Violet laser (405 nm) excitation; emission 600 nm.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases and antigen receptor signaling, crucial for T- and B-cell activation and cytokine receptor regulation. CD45 isoforms arise via alternative splicing and differ in extracellular domains. It also modulates integrin signaling, apoptosis, and various immune functions.
