Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and dendritic cells. CD14 binds LPS-LBP complexes with high affinity, facilitating immune recognition.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry using human peripheral blood cells (5 µL/test)
  • Recommended Cell Range: 10⁵–10⁸ cells/test

Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and is optimized for ultraviolet (355 nm) laser excitation.

Staining Recommendation:
When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and samples from light to prevent photo-induced oxidation.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) with minimal loss of brightness.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It acts as a multifunctional receptor for lipopolysaccharide (LPS) and interacts with LBP to mediate immune response. Soluble CD14 (sCD14) distinguishes structural differences in LPS and aids in neutralization of serum endotoxins. CD14 also binds apoptotic cells and influences allergic, inflammatory, and infectious processes. Alternative splicing yields multiple transcript variants of the same isoform.
