Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a mark for transcriptional activation, thereby acting as a corepressor. It contains a SWIRM domain, an FAD-binding motif, and an amine oxidase domain. LSD1 functions as part of histone deacetylase complexes and silences genes via histone demethylation. It oxidizes substrates through FAD to form imines that are hydrolyzed. LSD1 demethylates mono- and tri-methylated Lys-4 of histone H3 and may repress neuronal gene expression. It requires RCOR1/CoREST for activity on nucleosomes and may also demethylate Lys-9 of histone H3, influencing androgen receptor target gene expression. Mutations can lead to cleft palate, psychomotor retardation, and distinctive facial features.
