Product Specific Information
Annexins are a family of calcium-dependent phospholipid-binding proteins that bind to phosphatidylserine (PS) to identify apoptotic cells. In healthy cells, PS is located on the cytosolic side of the plasma membrane. During apoptosis, PS translocates to the extracellular side, which can be detected with fluorescently labeled Annexin V.
In early apoptosis, cells stained only with Annexin V (PI/7-AAD negative) are viable but undergoing apoptosis. In late apoptosis, loss of membrane integrity allows both Annexin V binding and uptake of PI or 7-AAD. Annexin V staining combined with PI or 7-AAD is widely used in flow cytometry to distinguish apoptotic stages.
Annexin V also functions as an anticoagulant protein by inhibiting prothrombin activation. Antibodies to Annexin V are associated with systemic lupus erythematosus (SLE), recurrent spontaneous abortions, and systemic sclerosis (SSc).
Purity: Annexin V is >89% pure as demonstrated by SDS-gel electrophoresis and reverse phase HPLC.
FITC labeling enables direct detection of PS by FACS analysis. Counterstaining with propidium iodide allows discrimination of apoptotic cells.
