Product Specific Information

Annexins are a family of calcium-dependent phospholipid-binding proteins that bind to phosphatidylserine (PS) to identify apoptotic cells. In healthy cells, PS is located on the cytosolic side of the plasma membrane. During apoptosis, PS translocates to the extracellular side, allowing detection with fluorescently labeled Annexin V.

In early apoptosis, cells are Annexin V positive but negative for propidium iodide (PI) or 7-AAD. In late apoptosis, loss of membrane integrity allows both Annexin V binding and uptake of PI or 7-AAD. Annexin V staining combined with PI or 7-AAD is widely used to distinguish apoptotic stages by flow cytometry.

Annexin V also functions as an anticoagulant protein in the blood coagulation cascade by inhibiting prothrombin activation. Antibodies to Annexin V are associated with systemic lupus erythematosus (SLE), recurrent spontaneous abortions, and systemic sclerosis (SSc).

Purity: Annexin V is >89% pure as demonstrated by SDS-PAGE and reverse-phase HPLC.
Biotin labeling: Enables direct detection of PS by FACS analysis using fluorescence-labeled Streptavidin. Counterstaining with PI allows discrimination of apoptotic cells.

Target Information

The gene encoding Annexin V belongs to the annexin family of calcium-dependent phospholipid-binding proteins, some of which are involved in membrane-related events in exocytotic and endocytotic pathways. Annexin V acts as a phospholipase A2 and protein kinase C inhibitory protein with calcium channel activity, playing roles in signal transduction, inflammation, growth, and differentiation.
