Target Information
Claudin-18은 폐와 위에서 특이적으로 발현되는 타이트 정션 단백질로, claudin-18 유전자의 대체 스플라이싱에 의해 생성된 이소폼이다. 폐 특이적 형태는 T/EBP/NKX2.1 전사인자에 의해 조절되는 하위 표적 유전자이다. 생쥐에서 C-말단 세포질 도메인이 결여된 스플라이스 변이가 존재하지만, 인간에서는 확인되지 않았다. 인간 Claudin-18은 생쥐 단백질과 88%의 아미노산 서열 유사성을 보인다. 면역조직화학 연구에서는 폐와 위 상피세포의 세포막에 완전하게 국한되어 있으며, 전자현미경 연구에서는 세포 간 접합부에 집중되어 있음이 확인되었다. 이러한 특징은 Claudin-18이 폐와 위의 타이트 정션 구조 및 기능에 중요한 역할을 할 가능성을 시사한다. 또한 Claudin-18 발현은 내이에서도 보고된 바 있다.
