Target Information

The protein encoded by this gene belongs to the SMAD family, which includes proteins similar to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways.

This protein mediates bone morphogenetic protein (BMP) signaling, influencing cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, this protein is phosphorylated and activated by the BMP receptor kinase, forming a complex with SMAD4 for transcriptional regulation. It is also a target for SMAD-specific E3 ubiquitin ligases (SMURF1, SMURF2), leading to ubiquitination and proteasomal degradation. Alternatively spliced transcript variants encoding the same protein have been observed.
