Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa, depending on phosphorylation. It is expressed on mature and most malignant B cells, a subpopulation of T lymphocytes, and follicular dendritic cells. CD20 regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation. It is associated with lipid rafts, and its association varies with extracellular triggers and BCR aggregation. CD20 remains on the cell surface after receptor ligation and is a key target for antibody-mediated B-cell depletion, as it is highly expressed on most B-cell malignancies and is not internalized or shed after treatment. Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
