Product Specific Information

PA1-973 detects proteasome 19S subunit S1 from human cells.
It has been successfully used in Western blot and immunohistochemistry procedures.
By Western blot, this antibody detects an approximately 105 kDa protein representing proteasome 19S subunit S1 from HeLa cell extract.
The immunizing peptide corresponds to amino acid residues 929–953 from human proteasome 19S subunit S1 protein.

Target Information

Proteolytic degradation is critical for maintaining appropriate levels of short-lived and regulatory proteins involved in various cellular processes such as metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle control, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most proteins in the eukaryotic cytosol and nucleus, while others are degraded via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.

The 26S proteasome is an ATP-dependent, multisubunit (~31) molecular complex (~2.5 MDa) consisting of a 20S proteolytic core and one or two 19S regulatory subunits.
The 19S subunits recognize ubiquitinated proteins and facilitate their unfolding and translocation into the 20S core.
An enzymatic cascade attaches multiple ubiquitin molecules to lysine residues of target proteins for degradation.
Defects in this pathway are linked to genetic disorders such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
