Target Information
PTEN은 다양한 암에서 돌연변이가 보고된 종양 억제 유전자로, phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase 활성을 가진 단백질을 암호화합니다. 이 단백질은 tensin 유사 도메인과 이중 특이성 단백질 타이로신 인산화효소와 유사한 촉매 도메인을 포함합니다. 대부분의 단백질 타이로신 인산화효소와 달리, PTEN은 인산화된 인지질 기질을 선호적으로 탈인산화합니다. PTEN은 세포 내 phosphatidylinositol-3,4,5-trisphosphate 수준을 음성 조절하여 AKT/PKB 신호 경로를 억제함으로써 종양 억제 기능을 수행합니다. 또한 PTEN은 성체 신경발생 과정에서 새로운 뉴런의 통합, 수상돌기 발달, 시냅스 형성 등을 조절하는 AKT-mTOR 신호 경로의 주요 조절자로 작용합니다. 지방 조직에서 인슐린 신호 및 포도당 대사의 음성 조절자로 작용할 수도 있습니다. 핵 내 단일 유비퀴틴화 형태는 더 높은 세포사멸 유도 능력을 가지며, 세포질 내 비유비퀴틴화 형태는 상대적으로 낮은 종양 억제 능력을 나타냅니다.
