Product Specific Information

The FF3399 monoclonal antibody recognizes human aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor mediating toxic effects of environmental contaminants such as PCB and TCDD. AHR also binds naturally occurring compounds and is localized in the cytoplasm with HSP90, p23, and XAP2/AIP/ARA9. Upon ligand binding, AHR translocates to the nucleus, forming a complex with ARNT to regulate genes like CYP1A1. It is expressed in many cell types, with highest levels in liver, and plays roles in Treg and Th17 cell differentiation.

  • Applications Tested: Pre-titrated and validated on Th17-polarized human peripheral blood cells using Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00)
  • Recommended Protocol: BestProtocols® “Protocol A” for cytoplasmic intracellular proteins
  • Usage: 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Fluorophore Characteristics:
PerCP-eFluor® 710 emits at 710 nm, excited by 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) with minimal brightness loss.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

AHR (Ah Receptor) belongs to the PAS domain family, including ARNT and Drosophila PER/SIM proteins. AHR binds specific DNA enhancer sequences to initiate CYP1A1 transcription. It regulates xenobiotic-metabolizing enzymes such as cytochrome P450. Ligands include polycyclic and halogenated aromatic hydrocarbons (e.g., TCDD). In inactive cells, AHR complexes with HSP90 in the cytoplasm; upon activation, it forms a nuclear heterodimer with ARNT. Associated diseases include eosinophilic fasciitis and seborrheic dermatitis.
