Target Information
Calcium-permeable channels such as TRPV6 participate in neurotransmission, muscle contraction, and exocytosis by providing calcium as an intracellular second messenger. Depending on tissue type, transcellular calcium transport may be regulated by vitamin D, parathyroid hormone (PTH), or calcitonin (CALCA).
TRPV6 functions as a calcium-selective cation channel involved in Ca²⁺ uptake in various tissues, including intestinal reabsorption. The channel is activated by low internal calcium levels and exhibits inward rectification. Inactivation occurs via both rapid Ca²⁺-dependent and slower Ca²⁺-calmodulin-dependent mechanisms, potentially regulated by phosphorylation.
In vitro, TRPV6 is slowly inhibited by Mg²⁺ in a voltage-independent manner. Heteromeric assembly with TRPV5 modifies channel properties, forming TRPV5–TRPV6 heteromultimeric concatemers that exhibit voltage-dependent gating.
TRPV6 interacts with TRPV5, S100A10, and the ANAX2–S100A10 heterotetramer, which are required for trafficking to the plasma membrane. It also interacts with BSPRY and calmodulin.
