Target Information
SMAD2 (MADH2, MAD2)는 세포 증식, 세포사멸, 분화 등의 다양한 세포 과정 조절에 관여합니다.
이 단백질은 TGF-β 수용체와 결합하여 핵으로 이동하며, 이는 TGF-β 신호전달의 중심 단계입니다.
ERK1에 의한 MH1 도메인 내 Thr8의 인산화는 SMAD2의 전사 활성화를 증가시키며, 이는 칼모듈린에 의해 음성 조절됩니다.
TGF-β 자극 시 SMAD2는 수용체에 의해 인산화되어 SARA와의 결합이 해리되고 SMAD4와 결합하여 핵으로 이동합니다.
SMAD4와의 복합체는 표적 유전자 프로모터에 결합하여 전사 억제 복합체를 형성합니다.
또한, SMAD2는 activin type 1 receptor kinase에 의해 인산화되어 activin 신호를 매개할 수 있습니다.
