Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at RT for 20 min.
Target Information
Spectrins are principal components of the cell membrane-cytoskeleton, composed of two alpha and two beta spectrin subunits.
SPTBN2 encodes spectrin beta non-erythrocytic 2 (beta-III spectrin), distinct from beta-II spectrin (SPTBN1).
SPTBN2 stabilizes the glutamate transporter EAAT4 at the plasma membrane surface, regulating glutamate signaling.
Mutations in SPTBN2 cause spinocerebellar ataxia type 5 (SCA5), characterized by neurodegeneration, progressive locomotor incoordination, dysarthria, and uncoordinated eye movements.
