Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was initially identified as a cell surface antigen of Hodgkin and Reed-Sternberg cells using monoclonal antibody Ki-1. Its ligand, CD30L (CD153), mediates effects such as cell proliferation, activation, differentiation, and apoptosis. CD30 plays a critical role in Hodgkin’s disease and other CD30+ lymphomas and acts as a costimulatory molecule in thymic negative selection.

CD30 expression is found in Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (including Burkitt’s lymphoma), virus-infected T and B cells, and activated normal T and B cells. In T cells, CD30 is expressed on subsets producing Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. The soluble form (sCD30) serves as a marker of Th2 immune response. TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T-cell proliferation, protecting against autoimmunity. Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
