Product Specific Information

For reconstitution, add 100 µL of distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 regulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, it comprises:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation correlates with tumor formation.
p53 plays a pivotal role in preventing proliferation of damaged cells and acts as an anticancer factor. It is expressed at low levels in normal cells but elevated in transformed cell lines, contributing to malignancy.
Mutations in the TP53 gene, common in various cancers, disrupt DNA binding and tumor suppressor function. Germline mutations are also associated with hereditary cancer syndromes such as Li-Fraumeni syndrome.
