Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. Upon stress, p53 activates transcription of specific genes such as p21waf1, leading to G1 or G2/M arrest.

p53 structure includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation is associated with tumor formation. p53 plays a vital role in preventing proliferation of damaged cells, functioning as an anticancer mechanism. Elevated p53 levels are linked to aging, oxidative stress, and deficits in long-term potentiation (LTP) related to learning and memory.

In normal cells, p53 is expressed at low levels, but in transformed cell lines, its expression increases significantly, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppression, and germline mutations are found in cancer-prone families such as Li-Fraumeni syndrome.
