Target Information
Human Rad18 (hRad18) is a homologue of the S. pombe Rad18. In yeast, Rad6 and Rad18 play a crucial role in post-replication repair, filling gaps in daughter strands during replication of damaged DNA. The Rad6–Rad18 interaction is critical at early steps of lesion bypass. Rad18 protein forms a tight complex with Rad6 through a conserved ring-finger motif, binding to damaged sites via its single-stranded DNA binding property. This leads to degradation of stalled replication components through the ubiquitin-conjugating activity of Rad6.
Human RAD18 maps to chromosome 3p24–25, a region often deleted in lung, breast, ovary, and testis cancers. The gene encodes a 495-amino-acid protein (~56 kDa) that is ubiquitously expressed in various human tissues.
