This peptide corresponds to 20 amino acid residues at the amino terminus of mouse ICAD.
PEP-0017 can be used as a blocking peptide with polyclonal antibody PA5-19889.
Target Information
ICAD is the inhibitor of caspase-3-activated DNase, which controls nuclear apoptosis.
It has two isoforms:
- ICAD-L/DFF45 (45 kDa): functional isoform
- ICAD-S/DFF35 (35 kDa): non-functional isoform
Although both ICAD-L and ICAD-S can bind and inhibit CAD, only ICAD-L is functional. ICAD is cleaved to allow caspase-activated DNase (CAD) to execute apoptotic DNA fragmentation. In non-apoptotic cells, CAD is complexed with ICAD. Activation occurs through caspase-3-mediated cleavage of ICAD at residues 117 and 224, producing three fragments released from CAD.
Beyond its DNase inhibitory role, ICAD acts as a CAD-specific folding chaperone. Recent studies suggest ICAD may be a potential target for restoring normal apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
