Target Information
CD22 (BL-CAM) is a type I integral membrane glycoprotein with a molecular weight of 130–140 kDa. It is expressed in both the cytoplasm and cell membrane of B-lymphocytes, appearing early in B-cell differentiation at a similar stage as CD19.
Unlike other B-cell markers, CD22 membrane expression is restricted to mature B cells (CD22⁺) and is absent in plasma cells (CD22⁻), making it useful in phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia and is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid, and the recognition site can be masked by cis interactions on the same cell surface. Ligand-induced tyrosine phosphorylation during immune responses suggests a regulatory role in B-cell receptor signaling. CD22 may act as both a positive and negative regulator by interacting with Src family kinases or recruiting cytoplasmic phosphatases that inhibit signal transduction.
