Product Specific Information

This antibody recognizes a non-polymorphic epitope on the mouse CD8 alpha chain. It blocks MHC I-dependent T cell responses in vitro and in vivo, and induces transplantation tolerance in combination with CD4 antibodies.

For FACS analysis, use 10 µL of the suggested working dilution to label 1×10^6 cells in 100 µL.


Target Information

Cluster of differentiation 8 (CD8), a type I transmembrane glycoprotein of the immunoglobulin family, plays a key role in signal transduction, T cell differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC-I molecules, while CD4 acts as a co-receptor for MHC-II molecules.

CD8α is essential for binding to MHC-I and is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
Ligation of MHC-I/peptide complexes by antigen-presenting cells (APCs) recruits Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs play a crucial role in pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
