Description:
The monoclonal antibody GAP.A3 reacts with the human major histocompatibility complex (MHC) class I molecule HLA-A3, a member of the HLA-A family. HLA-A3 exists as two subtypes (HLA-A3.1 and HLA-A3.2) differing at amino acid positions 152 and 156. MHC class I molecules are expressed on all nucleated cells and present intracellular peptides to cytotoxic T lymphocytes. HLA-A3 is expressed in 15–25% of cancer patients and binds the tumor-associated antigen hTERT.
Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.6 µg) per test. A test corresponds to staining a cell sample (10⁵–10⁸ cells) in a final volume of 100 µL.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
- Add 5 µL per sample to antibody cocktails or single-color controls (per 100 µL of cell sample).

Target Information
HLA-A belongs to the HLA class I heavy chain family. These heterodimers consist of a heavy chain and a light chain (β2-microglobulin). They are expressed on nearly all nucleated cells and play a key role in presenting peptides derived from the endoplasmic reticulum to the immune system. The heavy chain (~45 kDa) gene contains 8 exons encoding distinct structural domains.
Polymorphisms in exons 2 and 3 determine peptide-binding specificity and are used in transplantation typing. Hundreds of HLA-A alleles have been identified.
HLA and MHC antibodies are essential tools in Immunopeptidomics, enabling identification and characterization of neoantigens via LC–MS/MS.
