Product Specific Information

This OV9A2 monoclonal antibody reacts with human CD59 (Protectin), a GPI-linked membrane glycoprotein homologous to the murine Ly-6 superfamily. CD59 is expressed on all human lymphocytes, monocytes, granulocytes, erythrocytes, and also on non-hematopoietic cells such as endothelial cells and neurons.
By binding complement components C8 and C9, CD59 inhibits membrane attack complex (MAC) assembly and cytolytic activity. It also interacts with CD2 to modulate T cell adhesion and activation. Altered expression of CD55 and CD59 has been observed in systemic lupus erythematosus (SLE) patients.

Crossblocking studies indicate that OV9A2 binds the same epitope as MEM-43.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested for flow cytometry on normal human peripheral blood cells at 5 µL (0.5 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass.

Staining and Fixation:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.
Samples can be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) and stored up to 3 days at 2–8°C in the dark.

Target Information

CD59 (Protectin) is an 18–20 kDa GPI-anchored inhibitor of the complement membrane attack complex (MAC). It prevents cytolytic pore formation by binding complement C8 and/or C9, protecting cells from complement-mediated lysis.
CD59 also acts as a low-affinity ligand for CD2, contributing to T cell costimulation and signal transduction.
Mutations in CD59 cause CD59 deficiency, associated with hemolytic anemia, thrombosis, and cerebral infarction. Multiple alternatively spliced variants have been identified.
