Product Specific Information

Description:
The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported:
This antibody is validated for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Verify instrument compatibility before use.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein in the selectin family of adhesion receptors. It consists of:

• Aminoterminal C-type lectin binding domain
• Epidermal growth factor-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes including lymphocytes (except memory T cells), monocytes, and polymorphonuclear cells.
