Product Specific Information

The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against various tumor cells and plays a key role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. Secreted mainly by macrophages, it binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. TNF alpha forms multimers and shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Studies indicate its neuroprotective function and tumor necrosis activity. It plays a role in immune responses to infections and tumor cell necrosis and is implicated in autoimmune diseases, insulin resistance, and cancer.
