Product Specific Information

The MAb11 antibody reacts with human tumor necrosis factor-alpha (TNF alpha), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4+ T cells. TNF alpha has cytolytic activity against a range of tumor cells and plays a crucial role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form. The MAb11 antibody is a neutralizing antibody.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and protocol described in BestProtocols®.
• Use at 5 µL (0.25 µg) per test. One test = antibody amount required to stain a 100 µL cell sample (10⁵–10⁸ cells/test).

Handling Notes:

• Light sensitive: Protect from light during storage and staining.
• Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness.
• Excitation: 488–561 nm; Emission: 775 nm (Blue, Green, Yellow-Green lasers).
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is secreted mainly by macrophages and binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR receptors. TNF alpha regulates immune cell activity, apoptosis, lipid metabolism, and coagulation. It exists as multimers and shows a single 17 kDa band on SDS-PAGE under non-reducing conditions. It has roles in tumor necrosis, immune response to infection, and is implicated in autoimmune diseases, insulin resistance, and cancer.
