Target Information

MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted serine/threonine kinase belonging to the Ste20 kinase family. It acts as an upstream activator of the MEKK/JNK/SAPK signaling pathway, regulating T cell receptor (TCR) signaling and T cell proliferation through phosphorylation of the adaptor protein SLP-76. JNK/SAPK kinases are crucial for stress responses in mammalian cells. MAP4K1 is predominantly expressed in hematopoietic cells, including early progenitors, and specifically activates JNK1, enhancing AP-1-mediated transcription. Unlike PAK65, HPK1 lacks the Rac1/Cdc42-binding domain, indicating a distinct regulatory mechanism.
