Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases for signaling and receptor regulation. CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues on N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia (AML). It is found on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells, and acts as a marker for peripheral monocytes. CD33 helps distinguish myelogenous leukemia cells from lymphoid or erythroid leukemias. Dysfunction of CD33 is associated with certain malignancies such as gallbladder lymphoma and extracutaneous mastocytoma.
