Target Information

CD33는 sialic acid-binding immunoglobulin-like lectin (Siglec) 계열의 막관통 단백질로, ITIM(inhibitory motif)을 포함하여 SHP-1 및 SHP-2 단백질 인산화효소를 신호 복합체로 모집합니다. ITIM은 또한 세포 표면에서 수용체의 유비퀴틴 매개 제거에 관여합니다.

CD33는 골수계(myelomonocytic) 계열 세포에 발현되며, 세포 표면의 N- 및 O-glycan의 시알산 잔기와 결합합니다. 급성 골수성 백혈병(AML)의 치료 표적으로 연구되고 있으며, 골수 내 과립구 및 대식세포 전구세포에서 발현되지만, 다능성 줄기세포에서는 발현되지 않습니다. 또한 말초 단핵구(peripheral monocytes)에서도 발현되어, 백혈병 세포의 계통 구분에 유용한 표지자로 활용됩니다.

CD33 기능 이상은 담낭 림프종 및 피부 외 비만세포종과 관련이 있습니다.
