Description:
The 5D3 monoclonal antibody reacts with the extracellular portion of the human ABCG2 protein (also known as Bcrp1 and MXR). ABCG2, a member of the multi-drug resistance (MDR) family, is highly expressed in primitive side-population (SP) stem cells characterized by efflux of fluorescent dyes such as Rhodamine 123 and Hoechst 33342. In bone marrow, approximately 0.05% of cells show low fluorescence and are enriched for repopulating cells. SP cells expressing low or undetectable CD34 levels have been identified in multiple species, and ABCG2 expression appears highly conserved.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of ABCG2-transfected and peripheral blood cells. Recommended usage is 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration is recommended for specific assays.

ABCG2 (Breast Cancer Resistance Protein, BCRP1):
Also known as placenta-specific ABC transporter. Confers resistance to various chemotherapeutic agents including anthracyclines, mitoxantrone, bisantrene, and topotecan. Plays a protective role by removing toxins and regulating stem cell differentiation. Responsible for the side population (SP) phenotype and widely expressed in stem cells, placenta, liver, and intestine. May undergo N-linked glycosylation and dimerization in vivo.
