Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. Along with CD3γ and CD3δ, the ε chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.
Binding of 145-2C11 to TCR initiates intracellular signaling pathways leading to activation, proliferation, or apoptosis depending on experimental conditions. It is widely used as a phenotypic marker for mouse T cells.
Application Notes
- Reported for use in flow cytometric analysis
- Tested using mouse splenocytes
- Recommended usage: ≤1.0 µg/test (100 µL final volume)
- Cell count range: 10⁵–10⁸ cells/test
- Optimize antibody titration for best performance
Compatibility and Handling
- NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
- Each NovaFluor conjugate ships with CellBlox Blocking Buffer.
- Add 5 µL per sample when staining with NovaFluor conjugates.
- For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).
- Excitation: 496 nm; Emission: 511 nm; Laser: 488 nm (Blue)

Target Information
The CD3 subunit complex is essential for transducing antigen-recognition signals in T cells and regulating TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, all members of the immunoglobulin superfamily encoded on human chromosome 11.
- Plays a role in TCR-induced growth arrest, survival, and proliferation
- Expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes
- Absent on B and NK cells
- Decreased T lymphocytes can be observed in some autoimmune diseases
- Defects in CD3 genes are associated with CD3 immunodeficiency
