Target Information
The continued localization of p53 to the nucleus is essential for its tumor suppressor function. PARC (P53-associated parkin-like cytoplasmic protein), a large ubiquitin ligase, acts as a cytoplasmic anchor protein in p53-associated complexes. Inactivation of PARC leads to nuclear localization of p53 and activation of p53-dependent apoptosis, while overexpression promotes cytoplasmic sequestration of p53. Interestingly, PARC knockout mice are viable with no obvious phenotype, suggesting functional redundancy with other cullin family E3 ubiquitin ligases. p53 binding to PARC may also regulate PARC activity.
