Product Specific Information

Description:
The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed by developing and mature B cells but not plasma cells. CD20 is suggested to play a role in B-cell activation.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test (100 µL final volume).

Fluorochrome Information:
Brilliant Violet™ 711 (BV711) emits at 713 nm and is designed for cytometers equipped with a 405 nm violet laser. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV711 is not compatible with methanol-based fixation.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and malignant B cells, some T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 is associated with lipid rafts and remains at the cell surface after antibody ligation, making it a useful target for therapeutic B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
