Target Information
The mammalian Phospholipase Cγ (PLCγ) family includes two closely related proteins, PLCγ1 and PLCγ2. These isozymes share core structural domains and contain a unique set of additional domains: one PH domain, two SH2 domains, and one SH3 domain. Upon activation by receptor tyrosine kinases (RTKs) in response to extracellular stimuli such as hormones and growth factors, PLCγ catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to generate the second messenger molecules inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG).
