Target Information
STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. The STAT4 protein shares 52% similarity with STAT1 and 47% with STAT3. Functionally, STAT4 can be tyrosine phosphorylated by Jak1 or Jak2 and forms homo- or heterodimers with other STAT family members. Tyrosine-phosphorylated STAT4 binds the IFN-gamma activated site (GAS), and serine phosphorylation is required for maximal transcriptional activity.
STAT4 expression is mainly restricted to the thymus, spleen, and testis. It is activated by interleukin 12 (IL-12), which is essential for T-cell–independent induction of IFN-gamma and the development of Th1 responses necessary for defense against intracellular pathogens. STAT4-deficient mice show impaired IL-12–dependent Th1 development and enhanced Th2 differentiation. In viral infection, IFN-α/β–mediated activation of STAT4 is required for IFN-gamma production.
