Serclutamab is a biosimilar that targets EGFR.
EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1) is encoded by the EGFR gene located on chromosome 7 in humans.
EGFR belongs to the HER/ErbB family of proteins, which includes ERbB2, ERbB3, and ERbB4.
It is a transmembrane receptor that, upon ligand binding (e.g., EGF, TGFα), undergoes dimerization and autophosphorylation of tyrosine residues in its cytoplasmic domain.
EGFR activation triggers multiple signaling cascades such as Ras-ERK, PI3K-Akt, Jak-STAT, and PKC pathways, promoting cell growth and proliferation.
Phosphorylation at Y1086 enables GRB2 binding and MAPK pathway activation.
Activated EGFR is endocytosed and either degraded or recycled.
Mutations in EGFR are linked to lung cancer, and its overexpression correlates with poor prognosis in various tumors including those of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, and esophagus.
