Target Information
Caspase 8 (CASP8)는 cysteine-aspartic acid protease (caspase) 계열에 속하며, 세포사멸(apoptosis)의 실행 단계에서 핵심적인 역할을 합니다. Caspases는 비활성 전구체 형태로 존재하며, 활성화를 위해 내부 aspartic 잔기에서 절단되어 대형 및 소형 소단위체로 구성된 이합체 효소를 형성합니다.
Caspase 8은 Fas 및 다양한 apoptosis 자극에 의해 유도되는 programmed cell death에 관여하며, FADD와 상호작용하는 N말단 death effector domain을 가집니다. Huntington 병 환자의 뇌에서 Caspase 8이 불용성 분획에서 검출되어 신경퇴행성 질환과 관련이 있을 가능성이 제시되었습니다.
활성화된 Caspase 8은 Caspase 9 등 다른 하위 caspase를 활성화하여 세포사멸을 유도합니다. 또한 Jurkat 세포 및 편도(Tonsil)에서도 반응합니다. 과발현 시 apoptosis를 유도하며, ICE 계열 특이적 억제제로 차단될 수 있습니다. 다양한 스플라이싱 변이체가 보고되었으나, 모든 변이체의 full-length 서열은 확인되지 않았습니다.
