Despite differing sequences and substrates, they share the canonical α/β hydrolase fold and a catalytic triad (histidine, acid, nucleophile). Members of this superfamily are potential drug targets for conditions such as diabetes, Alzheimer’s disease, obesity, and blood clotting disorders.
The ABHD (Ab hydrolase domain containing) gene subfamily comprises 15 members, most of which utilize a serine nucleophile forming the G-X-S-X-G motif. Examples include:

- ABHD2: Involved in atherosclerosis development
- ABHD4: Participates in NAE synthesis pathways
- ABHD5: Mutations linked to Chanarin-Dorfman syndrome
