Product Specific Information
- 본 항체(700012)는 TPA 처리된 HeLa, NRK, MDCK 세포 용해물에서 phospho-ERK1/2 검출에 성공적으로 사용됨.
- 서열 상동성에 근거하여 소, 닭, 침팬지, 생쥐, 쥐, Xenopus, 제브라피시에서도 반응할 것으로 예측됨.
- 비환원 조건에서는 약 150 kDa의 완전 IgG 밴드로, 환원 시 약 25 kDa 경쇄 및 50 kDa 중쇄 밴드로 확인됨.
- 재조합 토끼 단클론 항체는 in vitro 발현 시스템을 통해 생산되며, 높은 특이성과 민감도, lot 간 일관성, 동물 유래 성분이 없는 제형, 그리고 넓은 면역 반응성을 제공함.
Target Information
ERK1 및 ERK2는 광범위하게 발현되며, 감수분열, 유사분열 및 분화된 세포의 기능 조절에 관여함.
성장인자, 사이토카인, 바이러스 감염, G 단백질 결합 수용체 리간드, 변형 인자 등 다양한 자극이 ERK1/2 경로를 활성화함.
성장인자가 수용체 티로신 키나아제에 결합하면 Ras가 Raf를 활성화하고, Raf는 MEK1/2를 인산화시켜 ERK1/2를 활성화함.
활성화된 ERK는 세포질 기질 단백질 또는 핵으로 이동하여 전사인자(c-Fos, Elk-1 등)를 인산화 및 활성화함.
