Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40, a member of the TNFR family, is expressed on B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype switching and, with IL-4, protects B cells from apoptosis while promoting proliferation.

Interaction of CD40 with its ligand CD154 (gp39) on T cells is critical for T-B cell crosstalk and immune regulation. The 5C3 antibody can be used for B cell activation in in vitro functional assays.

Target Information

CD40 is a single-chain glycoprotein and a member of the TNF/NGF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells and also found on some epithelial cells, carcinomas, and lymphoid dendritic cells.

CD40 is essential in mediating immune and inflammatory responses, including:

  • T cell-dependent immunoglobulin class switching
  • Memory B cell development
  • Germinal center formation

The transcription factor AKNA regulates CD40 expression, influencing cell interactions. CD40 interacts with adaptor protein TNFR2 for signal transduction and is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease. Two alternatively spliced transcript variants exist.
CD40 dysfunction is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
