Product Specific Information
The eB-h209 monoclonal antibody reacts with human CD209 (DC-SIGN), a 44 kDa type II transmembrane protein containing a C-type lectin binding domain. DC-SIGN binds ICAM-3, ICAM-2, and HIV virus, and is preferentially expressed on human dendritic cells. It is suggested to function as a receptor for HIV capture and transmission to T cells, supporting the primary immune response. The antibody was developed against a C-terminal peptide of human DC-SIGN.
- Applications Reported: Flow cytometric analysis
- Applications Tested: Tested by flow cytometry on cultured human dendritic and peripheral blood cells
- Recommended ≤ 1 µg per test (100 µL final volume)
- Cell number: 10⁵–10⁸ cells/test
- Optimize titration for best performance
- Filtration: 0.2 µm post-manufacturing filtered
Target Information
The CD209 gene encodes a transmembrane receptor (DC-SIGN) expressed on dendritic cells and macrophages. It plays a key role in the innate immune system by recognizing a wide range of pathogens including parasites and viruses. The protein has three main domains:
- N-terminal transmembrane domain
- Tandem-repeat neck domain
- C-type lectin carbohydrate recognition domain
The extracellular region functions as both a pathogen recognition and cell adhesion receptor. The neck domain allows homo-oligomerization, increasing binding avidity to multivalent ligands. Variations in the neck domain repeat number can affect ligand binding. CD209 is closely related to L-SIGN (GeneID 10332), differing in ligand-binding properties and tissue distribution. Alternative splicing results in multiple variants.
