Target Information

Arg3.1/ARC is a master regulator of synaptic plasticity that self-assembles into virion-like capsids encapsulating RNAs, mediating intercellular RNA transfer in the nervous system. ARC is released from neurons via extracellular vesicles, transferring ARC mRNA to new target cells where translation occurs in an activity-dependent manner.
ARC capsids are endocytosed and deliver mRNA into neuronal cytoplasm, functioning as a key regulator of synaptic plasticity, required for long-term potentiation (LTP), long-term depression (LTD), and long-term memory formation.
It promotes endocytosis of AMPA receptors (AMPARs) in response to synaptic activity, maintaining receptor levels through synaptic scaling. ARC also mediates synapse elimination in developing cerebellum and contributes to neuronal homeostasis.
Additionally, ARC participates in postsynaptic trafficking and processing of amyloid-beta (APP) and is expressed in skin-migratory dendritic cells, regulating dendritic cell migration and T-cell activation.
