Target Information
MMP (matrix metalloproteinase)는 결합 조직 성분을 분해할 수 있는 단백질분해 효소로, 공통적인 활성화 방식과 금속 결합 활성 부위의 보존된 아미노산 서열을 가지고 있으며, TIMPs(tissue inhibitors of metalloproteinases)에 의해 억제됩니다.
MMP와 TIMP는 혈관 신생(angiogenesis) 조절에 중요한 역할을 합니다.
MMP2는 631개의 아미노산으로 구성된 전구 효소(proenzyme)로 합성되며, 처음 80개의 아미노산 절단에 의해 활성화됩니다. 기본적으로 propeptide, catalytic, hemopexin 도메인을 포함합니다.
MMPs는 최소 18종 이상의 분비형 및 막결합형 zinc endopeptidase로 구성된 단백질군이며, 세포외기질(extracellular matrix)의 모든 구성 성분을 분해할 수 있습니다.
MMP2는 조직 재형성(tissue remodeling)에 관여하며, MMP-2 유전자 변이는 Winchester syndrome과 Nodulosis-Arthropathy-Osteolysis (NAO) syndrome과 관련이 있습니다.
두 가지 전사 변이(transcript variants)가 보고되어 있으며, 서로 다른 isoform을 암호화합니다.
