Target Information

WASP (Wiskott-Aldrich syndrome protein) and N-WASP are downstream effectors of Cdc42 involved in actin polymerization and cytoskeletal organization. The WASP family includes VASP and Mena, which accumulate at focal adhesions and regulate actin cytoskeleton dynamics. WAVE proteins, related to WASP family proteins, mediate actin reorganization downstream of Rho GTPases. WAVE1 and WAVE2 specifically regulate membrane ruffling by inducing actin filament clusters in response to Rac activation. These proteins cooperate with the Arp2/3 complex to promote actin filament formation. WAVE1 (also known as SCAR) is primarily expressed in the brain, while WAVE2 is broadly expressed, highest in leukocytes. WAVE3 forms a multiprotein complex linking receptor kinases and actin, all sharing a C-terminal verprolin homology domain for actin binding. This complex transduces signals related to cell shape, motility, and function.
