Target Information

DNA methylation, the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and gene expression regulation. The demethylation enzyme associated with a methyl-CpG-binding domain (MBD) can revert methylated cytosine bases to cytosines by cleaving the methyl group as methanol.
MeCP2 and MBD1 repress transcription by binding to methylated DNA, while MBD2 and MBD4 (also known as MED1) colocalize with methylated satellite DNA, mediating methylation signal functions.
MBD3 does not bind methylated DNA. MBD1–MBD4 are expressed in somatic tissues, though MBD1 and MBD2 expression is reduced or absent in embryonic stem cells lacking MeCP1 activity.
MBD4 shares homology with bacterial base excision repair DNA N-glycosylases/lyases and is known to be mutated in some microsatellite unstable tumors.
