Target Information
Caspase-activated DNase (CAD) cleaves chromosomal DNA during apoptosis. Apoptosis, a cell death process that removes toxic or unnecessary cells during mammalian development, can be induced by various stimuli such as death factors of the tumor necrosis factor family, UV irradiation, anticancer drugs, and survival factor deprivation.
The apoptotic process is characterized by shrinkage and fragmentation of cells and nuclei, and degradation of chromosomal DNA into nucleosomal units. CAD, activated by caspases, exists as a complex with its inhibitor ICAD (inhibitor of CAD). ICAD assists in proper folding of CAD and inhibits its DNase activity until cleaved by activated caspases. Once cleaved, CAD is released and enters the nucleus to cleave chromosomal DNA.
Apoptosis is regulated by the balance between pro- and anti-apoptotic proteins of the Bcl-2 family, heat shock proteins, and inhibitors of apoptosis proteins (IAPs). Fourteen members of the caspase family have been identified, seven of which mediate apoptosis.
